چکیده

The rabbit reticulocyte is capable of utilizing purines and their ribosyl derivatives for nucleoside triphosphate formation. Adenine, adenosine, hypoxanthine, and inosine are used extensively for the synthesis of adenosine triphosphate. Xanthine, guanine, and guanosine are utilized primarily for the synthesis of guanosine triphosphate. The inhibitor 6-diazo-S-oxo-L-norleucine inhibits the utilization of xanthine for guanosine triphosphate formation in rabbit erythrocytes, but does not affect its utilization for the synthesis of adenosine triphosphate. Similarly, the inhibitor interferes with the utilization of xanthine for guanosine triphosphate formation in human erythrocytes, without affecting its utilization for the synthesis of inosine monophosphate.

The utilization of purines and their ribosyl derivatives for the formation of adenosine triphosphate and guanosine triphosphate in the mature rabbit erythrocyte.

The finding that the purine portions of both adenosine triphosphate and guanosine triphosphate of the mature rabbit erythrocyte are not metabolically stable constituents of that cell, as is the heme of hemoglobin, suggested the possibility of a metabolic renewal of the nucleotides during the life of the cell (1, 2). The absence of any detectable utilization of sodium formate-Cl4 or of glycine-2...

Enzymatic deficiencies of purine nucleotide synthesis in the human erythrocyte.

In the development of the mature rabbit erythrocyte from the reticulocyte, the capacity for the synthesis de novo of the purine ring in vitro is lost (2, 3). The mature rabbit erythrocyte, however, does retain a portion of the biosynthetic pathway de novo, as evidenced by its utilization of 5-amino-1-ribosylWimidazolecarboxamide and sodium formate-Cl4 for the formation of inosinic acid (4). The...

The Biosynthesis of Purine Nucleotides de Nova in the Rabbit Reticulocyte in Vitro*

The mature rabbit erythrocyte has been shown to be incapable of synthesizing de novo the purine portions of adenosine triphosphate and of guanosine triphosphate, in vitro (2). It is able, however, to carry out the final steps of this pathway from sodium formate and 5-amino-1-ribosyl-4-imidazolecarboxamide, a finclmg which suggests that a portion of the over-all enzymatic sequence is lost during...

Interferon- Gamma- Inducible Guanosine Triphosphate Cyclohydrolase 1 (GTP-CH1) Pathway Is Associated with Frailty in Egyptian Elderly

Background: Chronic low-grade inflammation may be a cardinal pathophysiologic feature in the pathogenesis of frailty. Interferon-gamma (INF-γ) is an understudied proinflammatory cytokine in frailty that induces many inflammatory pathways including the guanosine triphosphate cyclohydrolase 1 (GTP-CH1) pathway. Our aim was to evaluate the GTP-CH1 pathway in Egyptian frail elderly subjects. ...
